Recommendation as being of special significance in the field by the Faculty of 1000


The article “The neck region of the C-type lectin DC-SIGN regulates its surface spatiotemporal organization and virus-binding capacity on antigen presenting cells.” recently published in The Journal of Biological Chemistry by Alessandra Cambi (Nanobiophysics group) and collaborators from the Institute of Photonic Sciences (Barcelona) has been recommended as being of special significance in its field by the Faculty of 1000 (F1000):

“DC-SIGN is a multipurpose pathogen receptor expressed in dendritic cells. This paper examines the functional role of nanoclusters, as visualized by stimulated emission depletion microscopy (STED) and near-field scanning microscopy, in pathogen binding -- focusing on viruses. The authors find the tandem repeat neck region is essential for nanocluster formation, so that when the neck region is deleted, the limited lateral mobility of DC-SIGN is markedly increased as assessed by quantum dot single particle tracking. Monte Carlo simulations of particles landing on a surface membrane reveal that nanoclusters enhance the binding of small virus particles when compared to a random distribution of receptors. Detailed studies such as this will further elucidate DC-SIGN biology and perhaps lead to new immunotherapeutic strategies.”
